Ketoprofen   100 mg

Mechanism of Action:

Ketoprofen shows anti-inflammatory and analgesic effects by inhibiting cyclooxygenase enzyme which is the main enzyme in conversion of arachidonic acid to prostaglandin, prostacycline and thromboxane.  Ketoprofen absorption is very fast after intramuscular injection. In horse, 99 percent of the drug is bound to the plasma albumin after absorption and then it is broadly distributed in different tissues. This drug reaches to articular and tendinous synovial fluid with proper concentration. Ketoprofen is metabolized via conjugation and its glucuronic acid ester is excreted via urine. Its half life in horse is 1 hour. If drug administration is repeated in animals with normal kidney and liver activity, it does not accumulate.

Various studies have demonstrated that Ketoprofen in comparison with NSAIDs such as phenylbutazone, has larger margin of safety so that administration of this drug in horse up to five times more than recommended dose has no drug poisoning.

Indications of use

Ketoprofen is used in cow and horse specially racing horse as a powerful analgesic, anti-inflammatory and antipyretic drug. Clinical studies have shown that ketoprofen has anti-inflammatory effects with powerful analgesic effects in different animals as follows:

Horse: Treatment of synovitis, arthritis, tenosynovitis, chondritis, osteochondritis, periostitis, laminitis, soft tissue inflammation and supportive treatment for sedation and improvement of postoperative inflammations. Ketoprofen is also used for reduction of pain and treatment of colic.

Cattle: Treatment of non infectious mastitis (physiological mastitis) or with antibiotics for treatment of infectious mastitis, in respiratory infections (pneumonia) in addition with antibiotics for reduction of inflammation and edema. It is also used for treatment of osteoarthritis and inflammation of soft tissues around the joints.

Dosage and route of administration

Horse: 2.2 mg/kg body weight per day through intramuscular or intravenous route for 5 days (1.1 ml Ketoprofen 10% per 50 kg body weight).

Cattle: 3 mg/kg body weight per day  through intramuscular or intravenous route for 3 days (1.5 ml Ketoprofen 10% per 50 kg body weight).

Withdrawal Time

Do not use meat of treated animals until 28 days after last injection and milk of treated animals until 7 days after last injection for human consumption.

Precautions

Do not use in human. Do not use in horses during racing period. Do not use in animals with hepatic or renal failures. Do not use through intra-articular route. Do not use if sedimentation occurs. Do not inject in joints.
